The Analogy Between Virus Spread and Lasing
From lasers, we know the following. Light can circulate in a laser resonator, where it experiences gain (amplification) and also some losses. The rate of change can be described with a simple differential equation:
where P is the circulating intracavity power, g the gain coefficient, l denotes the losses per round-trip and Trt is the round-trip time (often a few nanoseconds or even less). The gain depends on the amount of deposited excitation energy (e.g. through optical pumping) in the gain medium. As long as we have g < l, i.e., the net round-trip gain is negative, the laser is essentially off: although there is constantly some fluorescence light in the gain medium, it cannot be amplified to any significant level. If the gain exceeds the losses, however, i.e., we get a positive round-trip gain, the intracavity power will rise exponentially (proportional to exp((g − l) t / Trt)) until it starts saturating the gain, eventually limiting the further growth. Depending on the laser parameters, there may be some relaxation oscillations until a steady state can be reached (assuming continuous pumping).
The behavior of virus spread is somewhat similar. Infections are basically always caused by already infected people (apart from the initial infection, e.g. from some animal – usually a rare event). Therefore, the growth rate of the number of infected people is roughly proportional to that number itself. Although various details are substantially more complicated than those in lasers, we have a growth rate growing with the reached infection level, and thus potentially rapid growth over orders of magnitude. Also, we have a kind of threshold behavior similar to that in lasers: if the average number of people infected by one infected person (called the reproduction factor) is below 1, the infection will die out; if that number is larger, we will get exponential growth. So the reproduction factor in the epidemic exactly corresponds to the pump parameter of the laser.
So a crucial insight is that it makes a huge difference whether the reproduction factor is e.g. 1.1 or 0.9. While 1.1 means growth, potentially until the infection runs out of not-yet-infected people, 0.9 means a steady decline, never reaching a high level.
A very simple model illustrates essential features of a much more complicated system.
In detail, virus spread is really far more complicated to understand than lasers. For example, there is some time delay after infection where the infected person starts spreading viruses for some time. Also, the probability of further infections is not constant, but depends substantially on the region (e.g. the lifestyle of people) and on time (hopefully getting lower as people acknowledge the problem and learn to react properly). Nevertheless, the aspects of threshold behavior and exponential growth fully apply.
By the way, there are of course many other examples for exponential growth and threshold behavior in physics and technology. For example, a nuclear chain reaction essentially behaves like that. Here, exponential growth quickly leads to a nuclear explosion, releasing vast amounts of energy, if exponential growth is not stopped in time. On the other hand, the chain reaction in a nuclear reactor quickly dies out if it is just below criticality – similar to the vanishing epidemic when the reproduction factor gets below unity.

Respiratory droplets and contact transmission are most important routes of transmission of Coronavirus Disease (2019) (COVID-19), but do not fully account for the occurrence of all COVID-19 and there is other potential rout of transmission include aerosol transmission, body fluid routes and oral-fecal transmission.
Although the role of aerosol transmission of COVID-19 is not exactly clear, recent study focused on potential transmission of COVID-19 via aerosolization of particle of virus especially in closed environment in long time or in condition with high concentration of virus particle. Also viral ribonucleic acid (RNA) was found in anal swabs and in blood samples of infected patients.
Aerosols are liquid and solid particles suspended in the air. An aerosol with a diameter of 5 μm or less can remain airborne for a long period of time and enter the respiratory system. While most of the danger of lasers surgery in dermatology comes from the beam itself include direct eye injury, there are certain non-beam hazards include flammability, electrical hazards and airborne contaminants.
Airborne contaminants are associated with only certain wavelengths and applications of laser. It has demonstrated thermal disruption of viable human cells results in the release of carbon particles, virus, bacteria, deoxyribonucleic acid (DNA), and toxic gases. Regarding the outbreaks of COVID-19, in area with sustained community transmission, special consideration on laser safety with focus on potential biological hazards of moderate and high-power laser surgery is needed to reduce potential risks of laser airborne contaminants that provide more laser safety profiles and it is preferred that elective laser procedures postponed.

A father-son research team working from separate laboratory benches across the country has discovered a new use for lasers — zapping viruses out of blood. The technique, which holds promise for disinfecting blood for transfusions, uses a low-power laser beam with a pulse lasting just fractions of a second.
Johns Hopkins University student Shaw-Wei David Tsen says it was during a stroll in the park with his father that the idea was born. Tsen, an immunology researcher in the laboratory of T.C. Wu at Hopkins’ Kimmel Cancer Center, sought a new method to rid isolated blood of dangerous pathogens, including the viruses HIV and hepatitis C. He says current techniques using UV irradiation and radioisotopes can leave a trail of mutated or damaged blood components.
Using ultrasonic vibrations to destroy viruses was one possibility, but his father, Kong-Thon Tsen, a laser expert at Arizona State University, had a better idea: Lasers, unlike ultrasound, can penetrate energy-absorbing water surrounding the viruses and directly vibrate the pathogen itself.
The researchers aimed a low-power laser with a pulse lasting 100 femtoseconds (10-13 second) into glass tubes containing saline-diluted viruses that infect bacteria, also known as bacteriophages. The amount of infectious virus within each cube plummeted 100- to 1000-fold after the laser treatment. “I had to repeat the experiment several times to convince myself that the laser worked this well,” says the younger Tsen.
His laser is different from those emitting a continuous beam of visible light. “Our laser repeatedly sends a rapid pulse of light and then relaxes, allowing the solution surrounding the virus to cool off,” Tsen says. “This significantly reduces heat damage to normal blood components.”
Building on the idea that vibration wrecks a virus’ outer shell, the scientists found that their low-power laser selectively destroys viruses and spares normal human cells around them, while stronger beams kill almost everything.

We took a long time for this research, selecting each case individually. Was there a noticeably high number of “sudden and unexpected” deaths in sport and top-class sport until mid-2021? No official can (and will not) explain a possible accumulation since the start of gene vaccinations. We would like to start this report with the arguments of the other side, because it is always important to hear all sides. In 2016, the French medium Futura Santé complained of “numerous” heart deaths in football. It would affect 2 in 100,000 athletes per year in France – a total of 1,000 to 1,500 people. The New Statesman talks about 12 young people who die of sudden cardiac death while running – every week. The article was published in England in November 2018. In February 2019, the Austrian newspaper derStandard asked why cardiac deaths are “so common” in sport.

On the other hand, for example, there is an article in the German WAZ from June 13, 2021. There a cardiologist stated that sudden cardiac death rarely occurs in sport. Yes how now? Reliable statistics for comparison cannot be found. But in Wikipedia there are lists of athletes who died during the game. This list goes back to 1889 and at first glance is extensive – but in the end it turns out that about 5-8 such deaths were recorded even in “disaster years”. Many of these concern heart problems. The question is therefore difficult to answer without official and complete statistics:Are currently really more people dying “suddenly and unexpectedly” in sport, especially when it concerns professionals and top athletes? And is it because of the vaccination, as many critical minds suspect?
Over 75 known cases in the last 5 months
We have compiled a list of the known cases over the past few months. It is most likely incomplete. It includes many dead – and some who developed severe heart problems but fortunately survived. Noticeable: The youngest on the list was only 13 years old, many entries concern people under 20 years of age. From the gut one could say that there is much evidence of a noticeable accumulation in 2021. But why are the authorities not interested, why are there always excuses instead of strict examinations, precise autopsies?It may be that all of these people passed out dead or seriously ill on the field for “normal” reasons. But we certainly don’t know – and that is a criminal negligence of the rulers, whom we actually pay with billions in taxes to ensure that we have a good health system and that we only receive really safe and tested medication. Dirty money is piling up in Los Angeles. In the last three weeks, federal agents made three seizures that each netted more than $1 million in suspected drug proceeds.
The reason, according to the city’s top drug enforcement official: The coronavirus pandemic has slowed trade-based money laundering systems that drug trafficking groups use to repatriate profits and move Chinese capital into Southern California.
With storefronts closed, supply chains in disarray and the global economy in peril, these complex schemes are hobbled and cash is backing up in Los Angeles, Bill Bodner, special agent in charge of the Drug Enforcement Administration’s Los Angeles field division, said in an interview.
The recent million-dollar interceptions were reminiscent of seizures the DEA made before drug traffickers embraced trade-based money laundering, said Bodner, a 28-year agent.

I sell cannabis and cocaine to suppliers in the north of England. I have around 20 guys on the street, with approximately 200 regular customers. We have two main concerns now: sourcing drugs and getting enough money. We expect no more cocaine shipments from abroad for the next six weeks, so prices have shot up. I’m maintaining the same prices I’ve always charged but I’m concerned that, when stock begins to run low, people higher up the chain will charge more or cut the cocaine and decrease its quality. People are panicking – the amounts of cannabis they’re buying is ridiculous – so we are just dealing to regulars now. I’ve told my workers to be strict with what they sell and who to, but they aren’t changing their behaviour much, other than offering to post through letterboxes and accept bank transfers from trusted customers. People running out of money is a big concern, but we’ll always have the regular cokeheads who buy most days.

Clinical trial data contradicts ‘safe and effective’ government/industry mantra
Washington, DC, March 03, 2022 (GLOBE NEWSWIRE) — In a 55,000-page set of documents released on Tuesday, the U.S. Food and Drug Administration’s (FDA’s) Center for Biologics Evaluation and Research (CBER) is for the first time allowing the public to access data Pfizer submitted to FDA from its clinical trials in support of a COVID-19 vaccine license. This follows U.S. District Judge Mark T. Pittman’s decision on January 6 to deny the request from the FDA to suppress the data for the next 75 years which the agency claimed was necessary, in part, because of its “limited resources.”
A 38-page report included in the documents features an Appendix, “LIST OF ADVERSE EVENTS OF SPECIAL INTEREST,” that lists 1,291 different adverse events following vaccination. The list includes acute kidney injury, acute flaccid myelitis, anti-sperm antibody positive, brain stem embolism, brain stem thrombosis, cardiac arrest, cardiac failure, cardiac ventricular thrombosis, cardiogenic shock, central nervous system vasculitis, death neonatal, deep vein thrombosis, encephalitis brain stem, encephalitis hemorrhagic, frontal lobe epilepsy, foaming at mouth, epileptic psychosis, facial paralysis, fetal distress syndrome, gastrointestinal amyloidosis, generalized tonic-clonic seizure, Hashimoto’s encephalopathy, hepatic vascular thrombosis, herpes zoster reactivation, immune-mediated hepatitis, interstitial lung disease, jugular vein embolism, juvenile myoclonic epilepsy, liver injury, low birth weight, multisystem inflammatory syndrome in children, myocarditis, neonatal seizure, pancreatitis, pneumonia, stillbirth, tachycardia, temporal lobe epilepsy, testicular autoimmunity, thrombotic cerebral infarction, Type 1 diabetes mellitus, venous thrombosis neonatal, and vertebral artery thrombosis among 1,246 other medical conditions following vaccination.
“This is a bombshell,” said Children’s Health Defense (CHD) president and general counsel Mary Holland. “At least now we know why the FDA and Pfizer wanted to keep this data under wraps for 75 years. These findings should put an immediate end to the Pfizer COVID vaccines. The potential for serious harm is very clear, and those injured by the vaccines are prohibited from suing Pfizer for damages.”
The U.S. government has already purchased 50 million doses of the Pfizer vaccine intended for children under five years of age to be delivered by April 30, 2022 although the FDA has yet to grant an Emergency Use Authorization (EUA) for this age group. The risk of serious injury or death from COVID to healthy children is practically nil and so far, the vaccine is not effective when used in young children.
According to The Guardian, “Pfizer made nearly $37bn (£27bn) in sales from its Covid-19 vaccine last year – making it one of the most lucrative products in history – and has forecast another bumper year in 2022, with a big boost coming from its Covid-19 pill Paxlovid.” President Biden advertised Paxlovid in his State of the Union address on Tuesday, the same day the Pfizer data was released to the public. “We’re launching the ‘Test to Treat’ initiative so people can get tested at a pharmacy, and if they’re positive, receive antiviral pills on the spot at no cost,” Biden said during his speech.
From mid-December, 2020 through February 18, 2022, the U.S. government’s database, the Vaccine Adverse Events Reporting System (VAERS), has received 1,134,984 reports of adverse events, including 24,402 deaths, following COVID vaccination. Additionally, there have been 4,021 cases of myocarditis and pericarditis in the U.S. with 2,475 cases associated with Pfizer, 1,364 cases with Moderna and 171 cases with J&J’s COVID vaccine. These include 643 reports of myocarditis and pericarditis in children aged 12 to 17.
“It would be criminal to expose infants and young children to this extremely risky product,” said Holland. “VAERS data show the catastrophic health impacts the vaccine is having on millions of people, yet Pfizer and other vaccine makers are raking in billions of dollars with no fear of being held accountable for injuries and deaths from their vaccines.”

Vaccine failure dramatically worsens within 5-6 months after being vaccinated
One slide from the analysis reveals that so-called “breakthrough” infections — vaccine failures — increase with time, showing a near doubling of breakthrough infections among those vaccinated 5-6 months ago vs. those vaccinated only 3-4 months ago.
These data end at August 21st, 2021 but the trend does not appear to be flattening. As more data are added to this analysis each week, it seems almost certain that breakthrough infections rates will continue to rise over time in vaccinated individuals. We do not yet know what will happen in 9 months after vaccination, but these data show cause for serious concern.
Once the Delta variant took hold, 71% of COVID-19 “breakthrough” cases occurred among the fully vaccinated
As the following chart shows, 71% of COVID-19 “cases” were breakthrough cases (vaccine failures) once the Delta variant reached 90% spread across those infected.
Understand that the authors of this document state that those who are jabbed are not considered “vaccinated” until two weeks after they received the injections, which means that infections, hospitalizations and deaths which occurred from 0 – 14 days are ignored in this data set.
In reality, that means the percentage of “fully vaccinated” people responsible for breakthrough infections, hospitalizations and deaths is substantially higher than what is shown in these data. If they are claiming a 71% rate, it may in reality be more like 80% or even 90%, but we don’t know for sure because they are hiding all negative health outcomes for the first two weeks after the vaccines are administered (by claiming those people are “unvaccinated,” which is a deliberate deception being used to try to hide the harmful effects of vaccines).
Ethnic groups hit hardest: Native Americans, Hispanics and Blacks
Finally, a horrifying slide in the data set reveals that one of the highest risk factors for being hospitalized after being vaccinated is simply being of Native American descent. According to the data in this slide, Native Americans face around 50% higher odds of being hospitalized after being vaccinated, compared to other ethnic groups such as Whites.
Hispanics face a slightly lower risk which appears to be around 40% higher odds. Blacks face around 25% higher odds.
Why is this the case? The gain-of-function properties which were engineered into the SARS-CoV-2 biological weapon — via Fauci, Daszak and the NIH — target ACE2 receptors which exist in higher densities in targeted organ systems of many minority groups such as Native Americans, Hispanics and Blacks. This has led many observers to conclude that the covid spike protein — which is generated in the bodies of those who take mRNA vaccines — is a race-specific bioweapon designed to achieve depopulation of minority groups. Louis Farrakhan, leader of the Nation of Islam, has engaged in many efforts to bring this to the attention of his followers, for example. These data provided by the DoD / JAIC / Project Salus document shown here appear to support the plausibility of such theories.
Other factors that greatly increase a person’s odds of being hospitalized after receiving vaccinations include kidney failure (ESRD), morbid obesity, chronic liver disease or receiving chemotherapy.

The European Medicines Agency (EMA) has said it is investigating three new possible side effects that may be linked to mRNA Covid vaccines made by Pfizer and Moderna, after reports of allergic skin reactions and kidney disorders.
Europe’s medicines watchdog issued statements on Wednesday confirming that its Pharmacovigilance Risk Assessment Committee (PRAC) is looking into whether three medical conditions are connected to the administration of Covid jabs made by Pfizer and Moderna, after being made aware of a “small number” of incidences.
One of the three potential side effects being investigated is a skin disorder called erythema multiforme, an allergic reaction which manifests as reddish-purple skin lesions, sometimes around the mouth, eyes and other moist cavities on the body.
The two other conditions the PRAC is assessing for any link to the Pfizer and Moderna vaccines are related to the kidneys, with one disorder linked to inflammation of filters in the organ. The other, nephrotic syndrome, causes the kidneys to leak excess protein into the urine, which can then appear bloody or foamy. Symptoms of the disorder also include swollen eyelids, feet and abdomen, and fatigue. Some cases occurred in people with a history of kidney conditions.

A cytotoxic T cell (also known as TC, cytotoxic T lymphocyte, CTL, T-killer cell, cytolytic T cell, CD8+ T-cell or killer T cell) is a T lymphocyte (a type of white blood cell) that kills cancer cells, cells that are infected (particularly with viruses), or cells that are damaged in other ways.
Most cytotoxic T cells express T-cell receptors (TCRs) that can recognize a specific antigen. An antigen is a molecule capable of stimulating an immune response and is often produced by cancer cells or viruses. Antigens inside a cell are bound to class I MHC molecules, and brought to the surface of the cell by the class I MHC molecule, where they can be recognized by the T cell. If the TCR is specific for that antigen, it binds to the complex of the class I MHC molecule and the antigen, and the T cell destroys the cell.
In order for the TCR to bind to the class I MHC molecule, the former must be accompanied by a glycoprotein called CD8, which binds to the constant portion of the class I MHC molecule. Therefore, these T cells are called CD8+ T cells.
The affinity between CD8 and the MHC molecule keeps the TC cell and the target cell bound closely together during antigen-specific activation. CD8+ T cells are recognized as TC cells once they become activated and are generally classified as having a pre-defined cytotoxic role within the immune system. Summary
A common metabolic alteration in the tumor microenvironment (TME) is lipid accumulation, a feature associated with immune dysfunction. Here, we examined how CD8+ tumor infiltrating lymphocytes (TILs) respond to lipids within the TME. We found elevated concentrations of several classes of lipids in the TME and accumulation of these in CD8+ TILs. Lipid accumulation was associated with increased expression of CD36, a scavenger receptor for oxidized lipids, on CD8+ TILs, which also correlated with progressive T cell dysfunction. Cd36−/− T cells retained effector functions in the TME, as compared to WT counterparts. Mechanistically, CD36 promoted uptake of oxidized low-density lipoproteins (OxLDL) into T cells, and this induced lipid peroxidation and downstream activation of p38 kinase. Inhibition of p38 restored effector T cell functions in vitro, and resolution of lipid peroxidation by overexpression of glutathione peroxidase 4 restored functionalities in CD8+ TILs in vivo. Thus, an oxidized lipid-CD36 axis promotes intratumoral CD8+ T cell dysfunction and serves as a therapeutic avenue for immunotherapies.

Most of the recently risen coronavirus variants of interest or concern — delta, kappa, epsilon, iota and lambda — are universally carrying a mutation in amino acid L452 of the Spike protein that helps the virus bind to host cells.
Originally mutation L452R was discovered in early winter patient samples from California, first designated B.1.427 and B.1.429, and now known as the epsilon variant that underwent massive expansion across that state and is associated with many outbreaks.
In some of the samples, the researchers found another L452R-carrying virus variant that emerged more recently than the epsilon It is circulating in California and has shown up in other states states. This variant caused the only recorded case of COVID-19 in apes by infecting a gorilla troop at the San Diego Zoo in early January.
Unlike the epsilon variant with four more mutations in the Spike protein,in that newer variant, (from the lineage B.1.232), the L452R mutation is the only one in the Spike protein that latches the virus onto cells to create a pathway to inject its genetic materials. The Spike proteins are what give the coronavirus its studded, crown-like appearance.
The researchers determined that the L452R mutation alone triggered the emergence of the two California variants.
Additional analyses have shown that a dozen coronavirus lineages carrying L452R mutations have arisen around the world, including the globally expanded delta and delta plus variants that originated in Inda, as well as their cousin variant Kappa. Another variant of concern, lambda, that originated in Peru and Chili, is carrying mutation L452Q. Other mutations in those variants were also acquired but were not as ominpresent as the L452 mutations
A rise in mutational variants, particularly in this part of the virus, is of public health concern, because of the potential that they might make the pandemic coronavirus more infective, more virulent or more able to escape protective antibodies, the scientists conducting the study noted.
The findings seem to suggest that mutations in L452R, in and of themselves, offer significant adaptive value to the pandemic corona virus. From a virus evolution standpoint, positive selection for this mutation somehow became strong only in the past several months. It is not certain why this happened, but it could have occurred as the virus adapts to growing population immunity or the containment measures.

Background
There is an imbalance in the global distribution of COVID-19 vaccines. A sustainable increase in vaccine supply in LMICs must be achieved if the pandemic is to be contained. The manufacturing capacity for egg-based inactivated influenza vaccines (IIV) is among the largest in the world. Some of these facilities operate for less than half of the year and use embryonated eggs to manufacture more than a billion doses annually of affordable human vaccines.
To make use of these facilities, scientists have developed a COVID-19 vaccine for production in eggs. This vaccine is based on a Newcastle disease virus (NDV) expressing the ectodomain of a novel membrane-anchored, prefusion-stabilized SARS-CoV-2 spike protein construct (NDV-HXP-S). Between September and November 2020, manufacturers in Thailand, Vietnam, and Brazil modified their IIV manufacturing process and obtained high yields.
A new study
Scientists evaluated a range of vaccine doses (1 µg, 3 µg, 10 µg) having potency quantified as µg of virus envelope-anchored SARS-CoV-2 spike protein. Doses were evaluated in formulations with and without the TLR-9 agonist CpG1018 as a vaccine adjuvant.
Researchers observed that over four weeks after vaccination, all formulations were very well-tolerated with mild injection site pain and tenderness. Initial analysis of the clinical data raised no safety concerns, and the vaccine was strongly immunogenic in a formulation and dose-dependent manner. However, it was noted that the adjuvant benefit was uncertain as to the small sample size limited precision
Irrespective of the dose level, the vaccine-elicited neutralizing antibodies against two variants of concern (VoC), namely, B.1.351 and P.1. B.1.351 variant, is currently regarded as the worst-case example of immune evasion. Researchers observed that antibody titers decreased modestly against P.1 and more markedly against B.1.351, and this reduction depended on the assay used. This result was expected and in line with the range observed with sera from recipients of the vaccines BNT162b2 and mRNA-1273.
The NDV-HXP-S would be predicted to have neutralizing activity against the widely circulating delta (B.1.617.2) variant. The T cell response showed a bias towards a TH1 response, thereby alleviating concerns about enhanced disease associated with a TH2 response. The initial results are promising and suggest that the vaccine-induced T cell memory is capable of antiviral response.

Virophages are small, double-stranded DNA viral phages that require the co-infection of another virus. The co-infecting viruses are typically giant viruses. A newly discovered type of virus that spreads at the expense of other viruses, could be used to combat viral infections, say researchers.
Didier Raoult and colleagues from the University of the Mediterranean, France, say that the virus, called Sputnik, spreads by hijacking the replication machinery of the mamavirus – itself a new strain of the giant mimivirus.
The team says Sputnik is the first member of a new class they call “virophages” because of similarities with bacteriophages or phages – viruses that infect bacteria – and is the first time a virus has been seen to propagate at the expense of a viral host.
Research into phage therapy during the early 20th century was largely abandoned following the discovery of antibiotics.
Virus weapon?
Not only does Sputnik cut the spread of mamavirus in amoeba, Raoult’s analysis also shows it has looted genes from other viruses. This could help researchers understand the genetic evolution of harmful viruses, and potentially, use virophages to destroy them. However, the team is cautious.
“It’s too early to say we could use Sputnik as a weapon against big viruses or to modify them,” says co-author Bernard La Scola, also at the University of the Mediterranean. “But phages are used to modify bacteria, so why not?”
Sputnik resembles satellite viruses – such as the one that causes hepatitis D. These can only replicate in and infect their host if another virus is present. A key difference, though, is that Sputnik significantly reduces the viral load of the other virus.
“What is interesting here is that Sputnik is doing this at the expense of the bigger virus,” says Robin Weiss, of University College London.
However Geoffrey Smith, a virologist at Imperial College London, says this may not be surprising since both viruses are dependent upon the host cell for metabolites and will compete for them. He adds: “Bacteriophages replicate only in bacteria and that’s all they need, so the use of the phrase ‘virophage’ is inappropriate.”

The common analogy for CRISPR gene editing is that it works like molecular scissors, cutting out select sections of DNA. Stanley Qi, assistant professor of bioengineering at Stanford University, likes that analogy, but he thinks it’s time to reimagine CRISPR as a Swiss Army knife.
“CRISPR can be as simple as a cutter, or more advanced as a regulator, an editor, a labeler or imager. Many applications are emerging from this exciting field,” said Qi, who is also an assistant professor of chemical and systems biology in the Stanford School of Medicine and a Stanford ChEM-H institute scholar.
The many different CRISPR systems in use or being clinically tested for gene therapy of diseases in the eye, liver and brain, however, remain limited in their scope because they all suffer from the same flaw: they’re too large and, therefore, too hard to deliver into cells, tissues or living organisms.
In a paper published Sept. 3 in Molecular Cell, Qi and his collaborators announce what they believe is a major step forward for CRISPR: An efficient, multi-purpose, mini CRISPR system. Whereas the commonly used CRISPR systems—with names like Cas9 and Cas12a denoting various versions of CRISPR-associated (Cas) proteins—are made of about 1000 to 1500 amino acids, their “CasMINI” has 529.
The researchers confirmed in experiments that CasMINI could delete, activate and edit genetic code just like its beefier counterparts. Its smaller size means it should be easier to deliver into human cells and the human body, making it a potential tool for treating diverse ailments, including eye disease, organ degeneration and genetic diseases generally.

Abstract
We present a CRISPR-Cas based technique for deleting genes from the T7 bacteriophage genome. A DNA fragment encoding homologous arms to the target gene to be deleted is first cloned into a plasmid. The T7 phage is then propagated in Escherichia coli harboring this plasmid. During this propagation, some phage genomes undergo homologous recombination with the plasmid, thus deleting the targeted gene. To select for these genomes, the CRISPR-Cas system is used to cleave non-edited genomes, enabling isolation of the desired recombinant phages. This protocol allows seamless deletion of desired genes in a T7 phage, and can be expanded to other phages and other types of genetic manipulations as well.

To rapidly detect the presence of E. coli in drinking water, Cornell food scientists now can employ a bacteriophage – a genetically engineered virus – in a test used in hard-to-reach areas around the world.

Rather than sending water samples to laboratories and waiting days for results, this new test can be administered locally to obtain answers within hours, according to new research published by The Royal Society of Chemistry, August 2018.
“Drinking water contaminated with E. coli is a major public health concern,” said Sam Nugen ’99, Ph.D. ’08, Cornell associate professor of food science. “These phages can detect their host bacteria in sensitive situations, which means we can provide low-cost bacteria detection assays for field use – like food safety, animal health, bio-threat detection and medical diagnostics.”
The bacteriophage T7NLC carries a gene for an enzyme NLuc luciferase, similar to the protein that gives fireflies radiance. The luciferase is fused to a carbohydrate (sugar) binder, so that when the bacteriophage finds the E. coli in water, an infection starts, and the fusion enzyme is made. When released, the enzyme sticks to cellulose fibers and begins to luminesce.
After the bacteriophage binds to the E. coli, the phage shoots its DNA into the bacteria. “That is the beginning of the end for the E. coli,” said Nugen. The bacteriophage then lyses (breaks open) the bacterium, releasing the enzyme as well as additional phages to attack other E. coli.
Said Nugen: “This bacteriophage detects an indicator. If the test determines the presence of E. coli, then you should not be drinking the water, because it indicates possible fecal contamination.”

Bacteriophages represent a simple viral model of basic research with many possibilities for practical application. Due to their ability to infect and kill bacteria, their potential in the treatment of bacterial infection has been examined since their discovery. With advances in molecular biology and gene engineering, the phage application spectrum has been expanded to various medical and biotechnological fields. The construction of bacteriophages with an extended host range or longer viability in the mammalian bloodstream enhances their potential as an alternative to conventional antibiotic treatment. Insertion of active depolymerase genes to their genomes can enforce the biofilm disposal. They can also be engineered to transfer various compounds to the eukaryotic organisms and the bacterial culture, applicable for the vaccine, drug or gene delivery. Phage recombinant lytic enzymes can be applied as enzybiotics in medicine as well as in biotechnology for pathogen detection or programmed cell death in bacterial expression strains. Besides, modified bacteriophages with high specificity can be applied as bioprobes in detection tools to estimate the presence of pathogens in food industry, or utilized in the control of food-borne pathogens as part of the constructed phage-based biosorbents.

Significance
Understanding molecular events during virus assembly and genome packaging is important for understanding viral life cycles, and the functioning of other protein–nucleic acid machines. The model system developed for the thermophilic bacteriophage P23-45 offers advantages over other systems. Cryo-EM reconstructions reveal modifications to a canonical capsid protein fold, resulting in capsids that are abnormally large for this virus class. Structural information on the portal protein, through which the genome is packaged, demonstrates that the capsid influences the portal’s conformation. This has implications for understanding how processes inside and outside the capsid can be coordinated.

Abstract
Double-stranded DNA viruses, including bacteriophages and herpesviruses, package their genomes into preformed capsids, using ATP-driven motors. Seeking to advance structural and mechanistic understanding, we established in vitro packaging for a thermostable bacteriophage, P23-45 of Thermus thermophilus. Both the unexpanded procapsid and the expanded mature capsid can package DNA in the presence of packaging ATPase over the 20 °C to 70 °C temperature range, with optimum activity at 50 °C to 65 °C. Cryo-EM reconstructions for the mature and immature capsids at 3.7-Å and 4.4-Å resolution, respectively, reveal conformational changes during capsid expansion. Capsomer interactions in the expanded capsid are reinforced by formation of intersubunit ß-sheets with N-terminal segments of auxiliary protein trimers. Unexpectedly, the capsid has T=7 quasi-symmetry, despite the P23-45 genome being twice as large as those of known T=7 phages, in which the DNA is compacted to near-crystalline density. Our data explain this anomaly, showing how the canonical HK97 fold has adapted to double the volume of the capsid, while maintaining its structural integrity. Reconstructions of the procapsid and the expanded capsid defined the structure of the single vertex containing the portal protein. Together with a 1.95-Å resolution crystal structure of the portal protein and DNA packaging assays, these reconstructions indicate that capsid expansion affects the conformation of the portal protein, while still allowing DNA to be packaged. These observations suggest a mechanism by which structural events inside the capsid can be communicated to the outside.

Engineers at the University of California, Riverside, have altered a virus to arrange gold atoms into spheroids measuring a few nanometers in diameter. The finding could make production of some electronic components cheaper, easier, and faster.
“Nature has been assembling complex, highly organized nanostructures for millennia with precision and specificity far superior to the most advanced technological approaches,” said Elaine Haberer, a professor of electrical and computer engineering in UCR’s Marlan and Rosemary Bourns College of Engineering and senior author of the paper describing the breakthrough. “By understanding and harnessing these capabilities, this extraordinary nanoscale precision can be used to tailor and build highly advanced materials with previously unattainable performance.”
Viruses exist in a multitude of shapes and contain a wide range of receptors that bind to molecules. Genetically modifying the receptors to bind to ions of metals used in electronics causes these ions to “stick” to the virus, creating an object of the same size and shape. This procedure has been used to produce nanostructures used in battery electrodes, supercapacitors, sensors, biomedical tools, photocatalytic materials, and photovoltaics.
The virus’ natural shape has limited the range of possible metal shapes. Most viruses can change volume under different scenarios, but resist the dramatic alterations to their basic architecture that would permit other forms.
The M13 bacteriophage, however, is more flexible. Bacteriophages are a type of virus that infects bacteria, in this case, gram-negative bacteria, such as Escherichia coli, which is ubiquitous in the digestive tracts of humans and animals. M13 bacteriophages genetically modified to bind with gold are usually used to form long, golden nanowires.
Studies of the infection process of the M13 bacteriophage have shown the virus can be converted to a spheroid upon interaction with water and chloroform. Yet, until now, the M13 spheroid has been completely unexplored as a nanomaterial template.
Haberer’s group added a gold ion solution to M13 spheroids, creating gold nanobeads that are spiky and hollow.
“The novelty of our work lies in the optimization and demonstration of a viral template, which overcomes the geometric constraints associated with most other viruses,” Haberer said. “We used a simple conversion process to make the M13 virus synthesize inorganic spherical nanoshells tens of nanometers in diameter, as well as nanowires nearly 1 micron in length.”
The researchers are using the gold nanobeads to remove pollutants from wastewater through enhanced photocatalytic behavior.
The work enhances the utility of the M13 bacteriophage as a scaffold for nanomaterial synthesis. The researchers believe the M13 bacteriophage template transformation scheme described in the paper can be extended to related bacteriophages.
The paper, “M13 bacteriophage spheroids as scaffolds for directed synthesis of spiky gold nanostructures,” was published in the July 21 issue of Nanoscale. Other authors, all based at UCR, include Tam-Triet Ngo-Duc, a doctoral student in materials science and engineering; Joshua M. Plank, a doctoral student in electrical and computer engineering; Gongde Chen, a doctoral student in chemical and environmental engineering; Reed E. S. Harrison, a doctoral student in bioengineering; Dimitrios Morikis, a professor of bioengineering; and Haizhou Liu, a professor of chemical and environmental engineering.

Abstract
We genetically modified tobacco mosaic virus (TMV) to surface display a characterized peptide with potent metal ion binding and reducing capacity (MBP TMV), and demonstrate that unlike wild type TMV, this construct can lead to the formation of discrete 10–40 nm gold nanoparticles when mixed with 3 mM potassium tetrachloroaurate. Using a variety of analytical physicochemical approaches it was found that these nanoparticles were crystalline in nature and stable. Given that the MBP TMV can produce metal nanomaterials in the absence of chemical reductants, it may have utility in the green production of metal nanomaterials.

Sialic acids are a class of alpha-keto acid sugars with a nine-carbon backbone. The term “sialic acid” (from the Greek for saliva, σίαλον – síalon) was first introduced by Swedish biochemist Gunnar Blix in 1952. The most common member of this group is N-acetylneuraminic acid (Neu5Ac or NANA) found in animals and some prokaryotes.
Sialic acids are found widely distributed in animal tissues and related forms are found to a lesser extent in other organisms like in some micro-algae, bacteria and archaea. Sialic acids are commonly part of glycoproteins, glycolipids or gangliosides, where they decorate the end of sugar chains at the surface of cells or soluble proteins. However, sialic acids have been also observed in Drosophila embryos and other insects. Generally, plants seem not to contain or display sialic acids.
In humans the brain has the highest sialic acid content, where these acids play an important role in neural transmission and ganglioside structure in synaptogenesis. More than 50 kinds of sialic acid are known, all of which can be obtained from a molecule of neuraminic acid by substituting its amino group of one of its hydroxyl groups. In general, the amino group bears either an acetyl or a glycolyl group, but other modifications have been described. These modifications along with linkages have shown to be tissue specific and developmentally regulated expressions, so some of them are only found on certain types of glycoconjugates in specific cells. The hydroxyl substituents may vary considerably; acetyl, lactyl, methyl, sulfate, and phosphate groups have been found.

Fool the novel coronavirus once and it can’t cause infection of cells, new research suggests.
Scientists have developed protein fragments – called peptides – that fit snugly into a groove on the SARS-CoV-2 Spike protein that it would normally use to access a host cell. These peptides effectively trick the virus into “shaking hands” with a replica rather than with the actual protein on a cell’s surface that lets the virus in.
Previous research has determined that the novel coronavirus binds to a receptor protein on a target cell’s surface called ACE2. This receptor is located on certain types of human cells in the lung and nasal cavity, providing SARS-CoV-2 many access points to infect the body.
For this work, Ohio State University scientists designed and tested peptides that resemble ACE2 enough to convince the coronavirus to bind to them, an action that blocks the virus’s ability to actually get inside the cell.

New Biochemical Clues in Cell Receptors Help Explain How Coronavirus May Hijack Human Cells

The SARS-CoV-2 virus may enter and replicate in human cells by exploiting newly-identified sequences within cell receptors, according to work from two teams of scientists.
The findings from both groups paint a more complete portrait of the various cellular processes that SARS-CoV-2 targets to not only enter cells, but to then multiply and spread. The results also hint that the sequences could potentially serve as targets for new therapies for patients with COVID-19, although validation in cells and animal models is needed.
Scientists know that SARS-CoV-2 binds the ACE2 receptor on the surface of human cells, after which it enters the cell through a process known as endocytosis. Research has suggested that the virus may hijack or interfere with other processes such as cellular housekeeping (autophagy) by targeting other receptors called integrins. However, not much is known about exactly how the virus takes advantage of integrins on the biochemical level.
Analyzing the Eukaryotic Linear Motif database, Bálint Mészáros and colleagues discovered that ACE2 and various integrins contained several short linear motifs (SLiMs ) — small amino acid sequences — that they predicted play a role in endocytosis and autophagy. The scientists then compiled a list of currently used experimental treatments and approved drugs that can target the interactions between SARS-CoV-2 and its target SLiMs.
Separately, Johanna Kliche and colleagues performed molecular tests to see whether these SLiMs interacted with proteins that contribute to autophagy and endocytosis. The team found that two SLiMs in ACE2 bound to the endocytosis-related proteins SNX27 and SHANK, and one SLiM in the integrin β3 bound to two proteins involved in autophagy.
In addition to providing a resource for repurposing drugs for SARS-CoV-2, Mészáros et al. say their prediction methods could help identify similar under-the-radar SLiMs that assist with the replication of other viruses that cause disease.

A Chinese Communist Party military scientist who got funding from the National Institutes of Health filed a patent for a COVID-19 vaccine in February last year — raising fears the shot was being studied even before the pandemic became public, according to a new report.
Zhou Yusen, a decorated military scientist for the People’s Liberation Army (PLA) who worked alongside the Wuhan Institute of Virology as well as US scientists, filed a patent on Feb. 24 2020, according to documents obtained by The Australian.
The patent — lodged by the “Institute of Military Medicine, Academy of Military Sciences of the PLA” — was filed just five weeks after China admitted there was human-to-human transmission of the virus, and months before Zhou died under mysterious circumstances, the report noted.
“This is something we have never seen achieved before, raising the question of whether this work may have started much earlier,” Prof. Nikolai Petrovsky from Flinders University told the paper.
Adding to the intrigue, Zhou later died under mysterious circumstances in May last year — something being looked into as part of the international investigation ordered by President Biden, the paper insisted.
Despite being an award-winning military scientist, there were no reports or tributes, with him just listed as “deceased” in a Chinese media report in July and a December scientific paper…

These postulates are scientific steps used to prove whether a virus exists and has a one-to-one relationship with a specific disease. We showed that to date no one has proven SARS-CoV-2 causes a discrete illness matching the characteristics of all the people who ostensibly died from COVID-19. Nor has the virus has been isolated, reproduced and then shown to cause this discrete illness.
In addition, in a June 27 Off-Guardian article two more journalists, Torsten Engelbrecht and Konstantin Demeter, added to the evidence that “the existence of SARS-CoV-2 RNA is based on faith, not fact.”
The pair also confirmed “there is no scientific proof that those RNA sequences [deemed to match that of the novel coronavirus] are the causative agent of what is called COVID-19.”

This article may be misleading. There are specific monoclonal antibodies available. (https://www.ncbi.nlm.nih.gov/pmc/articles/PMC7048180/). Many companies supply them online globally. Maybe this pathologists is really implying that most people that die, actually pass from other conditions that pathologists can confirm are the cause of death.

A commonly-used pesticide could be partially responsible for the global obesity epidemic, says a study led by McMaster University scientists.
Researchers discovered that chlorpyrifos, which is banned for use on foods in Canada but widely sprayed on fruits and vegetables in many other parts of the world, slows down the burning of calories in the brown adipose tissue of mice. Reducing this burning of calories, a process known as diet-induced thermogenesis, causes the body to store these extra calories, promoting obesity.
Scientists made the discovery after studying 34 commonly used pesticides and herbicides in brown fat cells and testing the effects of chlorpyrifos in mice fed high calorie diets. Their findings were published in Nature Communications and could have important implications for public health.

Coronaviruses are found in a diverse array of bat and bird species, which are believed to act as natural hosts. Molecular clock dating analyses of coronaviruses suggest that the most recent common ancestor of these viruses existed around 10,000 years ago. This relatively young age is in sharp contrast to the ancient evolutionary history of their putative natural hosts, which began diversifying tens of millions of years ago. Here, we attempted to resolve this discrepancy by applying more realistic evolutionary models that have previously revealed the ancient evolutionary history of other RNA viruses. By explicitly modeling variation in the strength of natural selection over time and thereby improving the modeling of substitution saturation, we found that the time to the most recent ancestor common for all coronaviruses is likely far greater (millions of years) than the previously inferred range.

A new analysis of 139 instances where an animal virus has spread to humans shows these outbreaks are a human problem with a human cause

COVID-19 is all on us.
Up to 75 per cent of human infectious diseases that have recently emerged are known or are suspected to have originated from animals. This includes the coronavirus disease 2019 (COVID-19), which is caused by the 2019 novel coronavirus (SARS-CoV-2) and is now pandemic.
Our team from the University of California, Davis and the University of Melbourne, studied 139 different zoonotic viruses – where the virus has spread from non-human animals to humans.
Our study published in Proceedings of the Royal Society B, identified the animal hosts as well as the drivers of transmission – the factors that make it possible for a virus to spread.
From looking at the factors that lead to a virus jumping from a non-human animal population to us, one thing became clear: though animals may be the original natural host for these viruses, the drivers of a spillover are all too human.
Why does a virus spillover to humans?
Coronaviruses circulate in mammals and birds.
Bats are suspected as the original evolutionary source for the coronavirus that spilled over causing COVID-19 in humans.
Researchers have also found that SARS-CoV-2-related coronaviruses in pangolins — or scaly anteaters— share genetic material with the virus causing COVID-19, but we won’t know for certain which species were involved in spillover of this virus until more research is done.
Zoonotic coronaviruses appear to be very good at adapting to new host species.
Three coronaviruses that are zoonotic cause severe, potentially fatal, respiratory infections in humans; Severe acute respiratory syndrome (SARS-CoV), Middle East respiratory syndrome (MERS-CoV), and the newly emerged COVID-19.
SARS-CoV for example, in scientific terms, was found to have higher than average host plasticity. That is, it has a diverse host range, having been detected in a large number of animal taxonomic orders and species.
Viruses with high host plasticity are the most likely to become capable of human-to-human transmission and as we’ve seen with COVID-19, global spread.
Our previous research confirms these viruses are transmitted to us because we facilitate the mixing of diverse animal species.
This includes at live animal markets that sell wildlife, which have been attributed to the emergence of COVID-19, as well as other activities, such as keeping wild animals in sanctuaries or as pets.
Activities that increase our opportunity for animal-human interactions also likely facilitate zoonotic disease transmission.
The most frequent locations of transmission were in and around human dwellings and in agricultural fields, as well as at those interfaces with occupational exposure to animals: like. hunters, laboratory workers, veterinarians, researchers, wildlife management, as well as zoo and sanctuary staff.
Most of these high-risk interfaces have been driven by factors related to global animal species abundance.
More animals with less biodiversitY means more problems
Our new research found zoonotic viruses have historically come from animal species that have increased in abundance and particularly, have adapted to human-dominated landscapes and even expanded their range because of this.
Resilient wild animal species include rodents, primates, bats, even-toed ungulates (hoofed animals) and carnivores, which host the highest proportion of zoonotic viruses.
The presence of rodents and bats in that line-up may not surprise you.
Rodents – the house mouse and black rat – were the only wild animals among the top 10 species with the highest number of detected zoonotic viruses.
Both of these rodent species are considered invasive in most regions of the world, commonly inhabit domestic and peri-domestic structures, are used in laboratory studies and kept as pets.
However, when combining all species information into taxonomic order, primates and bats were more likely to harbour zoonotic viruses compared with all of the other orders.
Resilience to habitat changes brought on by humans isn’t the only factor that increases zoonotic virus spillover. Certain causes of decline in wildlife populations have also facilitated spillover of animal viruses to humans.
Importantly, among threatened wildlife species, those with population reductions due to exploitation and loss of habitat, this includes some species of primates and flying foxes – likely due to human encroachment – shared more viruses with humans.
Reducing the risk FOR another COVID-19
This means that in situations like the one we now face with COVID-19, we mustn’t blame the animals.
Not for moral reasons, for practical ones.
Domesticated animals kept as pets or on farms host more zoonotic viruses than wild species overall, but are less likely to be implicated in spillover of recently emerged novel viruses to humans.
Another difference is that for viruses hosted by domestic animals, such as rabies in dogs, we have fortunately developed a vaccine.
